The Advanced Gene Targeting Facility is a service offered by the Institute of Experimental Immunology and is aimed at providing researchers the possibility to generate gene-targeted mouse strains of their choice.
We provide service and support on different levels.
We currently offer to perform whole targeting experiments in different embryonic stem cell (ES) lines in various backgrounds such as C57BL/6, 129/Ola, F1:C57BL/6X129/SV, and BALB/c.
We expand gene trapped or gene deficient ES cell lines from the International Gene Trap and Knockout Consortia and prepare the cells for morula aggregation.
The morula aggregation service for the generation of chimeric mice is provided jointly with the TRT lab (Pawel Pelczar pawel.pelczar@ltk.uzh.ch (044-255-3737)) of the Institute of Animal Sciences (Labortierkunde, LTK). Please register for morula aggregation on the webpage of the TRT lab.
Our service also offers the establishment and use of Southern blot screens, DNA-preparation in 96-well format for PCR-screen of targeted ES cells.
We support researchers in the design of a targeting strategy, the cloning of targeting vectors for conditional mutagenesis, screening of the ES cells, and the best breeding strategy. Our laboratory offers support for the generation of BAC trangenic mouse lines (Cre, CreERt2, lacZ, tdTomato, GFP, channelrhodopsin and more) that are made by oocyte injection with the TRT lab.
